Ribonucleases (RNase) are present in all cell types and organisms, and their ubiquitous nature can cause numerous problems in the laboratory. Contamination with even a small amount of RNase in an RNA sample is enough to completely destroy the RNA. RiboShield® RNase Inhibitor is a recombinant human placental protein capable of blocking the activity of multiple types of ribonuclease, including RNase A, B, and C, to reliably protect your RNA from RNase degradation.
RNase Inhibitor with Superior RNA Protection
RiboShield® RNase Inhibitor is designed for RNA-sensitive applications including RNA extraction, RT-qPCR, cDNA synthesis, and RNA sequencing, to protect your RNA from degradation, yielding higher and better performance.
The inhibitor non-covalently binds to RNases at a 1:1 ratio and possesses very rapid binding kinetics, ensuring immediate protection for your RNA. RiboShield® RNase Inhibitor has been tested in RT-qPCR and offers superior RNA protection when compared to leading competitor products (see Figure 1).
Stable at Higher Temperatures
RiboShield® RNase Inhibitor is effective under many reaction conditions and can sustain RNase A inhibition at temperatures up to 65°C for at least 30 minutes (see Figure 2). Furthermore, RiboShield® does not contain cysteine residues known to be associated with oxidation sensitivity in the human placental protein version1. This results in an RNase inhibitor molecule that is not only temperature-stable but also shows enhanced resistance to oxidative stress.
Versatile RNase Inhibition Applications
The inhibitor can be used to block the activity of a broad range of ribonucleases, including eukaryotic neutral RNases (RNase A, B, and C). It does not inhibit RNase T1, T2, U1, U2, CL3, RNase I, and H. The inhibitor does not interfere with other enzymes such as reverse transcriptases, RNA polymerases, or Taq DNA polymerase, making it compatible with many RNA-related enzymatic reactions.
RiboShield® RNase Inhibitor has a molecular weight of 50 kDa and is purified from High Five insect cells expressing a modified human placental gene. The inhibitor non-covalently binds to RNases in a 1:1 ratio, and has a Ki value of approximately 10–14 M when binding RNase A1. Moreover, the very rapid binding kinetics with RNases ensures immediate protection for your RNA.

How much RiboShield® should I add to RNA samples for storage?
For storage of samples, we recommend using RiboShield® RNase Inhibitor at a 100x solution (i.e., the concentration of RNase inhibitor in the storage solution should be 0.4 U/μL). This final concentration can be exceeded without adversely affecting downstream enzymatic reactions, but it is generally unnecessary unless RNase contamination is suspected.
How to use RiboShield® RNase Inhibitor?
All PCRBIO products for one-step and two-step RT-PCR as well as for cDNA synthesis already contain RNase inhibitor, so there is no need to add RiboShield® RNase Inhibitor when using these products.
If using with other kits, we recommend adding 1µL (40U) RiboShield® RNase Inhibitor to a 20µL reaction mix. In cases where the RNA source is known to be highly contaminated with RNases, titration may be required to verify complete protection of ribonucleic acid.
What is RiboShield® RNase Inhibitor used for?
RiboShield® RNase Inhibitor is designed for use in RNA-sensitive applications such as RT-qPCR, cDNA synthesis, and RNA-seq, where the presence of even a small amount of RNase can harm RNA quality and experimental results. Additionally, it can be added to extracted RNA to preserve RNA quality in high-risk areas for RNase contamination or in RNA extraction solutions like those used in the study by Pereira et al., 2010, to enhance RNA integrity preservation as you proceed through your workflow.
